In need of Fully Customize Panel Arrangement

I am attempting to customize the layout with combination of columns and rows as shown in the image below. At the moment, I cannot achieve this using the layout settings which limits the ability to have 2 panels, 3 panels, 3 columns, and 4 panels.

Is there a way that this can be accomplish and persist?
